Protocols for Dissecting, Trimming, and Embedding Rodent Tissues
Rodent Stomach (modified from Dr. Nobuyuki Ito et al., by Dr. Seiko Tamano, Nagoya City University Medical School, Nagoya, Japan)
1- Resection of Fixation
Ligate the resected stomach at the portion of the cardiac esophagus and duodenum, inflate it by the injection of fixative, and place it in a small cup filled with the same fixative for five minutes. Open the stomach by cutting the greater curvature with a sharp razor blade, flatten and staple it between folded filter paper on which the histology number is written. It must remain fixed until trimming.
2- Trimming
Cut four sections from the fundic regions and two sections from the pyloric regions perpendicularly. As illustrated below, put trimmed pieces between foam pads in a cassette.
If stomach masses are present, trim as indicated, but place 1-2 slices (whichever fits) per cassette. Identify section number and orientation in the cassette.
3- Paraffin Embedding
